چکیده

The goal of the present study was to investigate production of cellulase in low cost medium by thermotolerant yeast. After screening, an efficient yeast isolate having capability of C1 (exo-gluconase) and Cx (endogluconase) production was isolated and designated as strain R-1. Maximum enzyme production was achieved at 50 °C, pH 5.5 in the medium containing bagasse powder 4% (w/v), and ammonium sulphate 0.1% (w/v) after 72 hours of incubation. The composition containing bagasse powder, 4% (w/v); ammonium sulphate, 0.5 % (w/v); and glucose, 0.5% (w/v) achieved better production after complete medium optimization. The yeast isolate was able to tolerate wide ranges of temperature, pH, and substrate concentration for higher enzyme production. The isolated yeast was able to produce C1 (exo-gluconase) and Cx (endo-gluconase) enzymes in appropriate concentrations on a crude cellulosic substrate. Therefore, yeast may be used to power alcohol production.

Optimization of the Cellulase Free Xylanase Production by Immobilized Bacillus Pumilus

Background: The extracellular xylanase secreted by microorganisms is a hydrolytic enzyme, which arbitrarily cleaves the β-1, 4 backbone of the polysaccharide xylan; an enzyme used in the food processing, bio-pulping and bio-bleaching. The commercial production of the xylanase is limited because of a higher cost involvement, which can be overcome by the cost-effective production...

Optimization of Culture Conditions and Medium Components for Carboxymethyl Cellulase (CMCase) Production by a Novel Basidiomycete Strain Peniophora sp. NDVN01

Background: Cellulytic enzymes including carboxymethyl cellulases play the key role in hydrolysis of cellulose, a huge organic carbon reservoir on earth, into monomeric sugars and their eventual conversion into valuable chemicals and energy sources. Objectives: In this study, we described the identification of a basidiomycete isolate NDVN01 and optimization of culture conditions and medium com...

Bioconversion of paper sludge to biofuel by simultaneous saccharification and fermentation using a cellulase of paper sludge origin and thermotolerant Saccharomyces cerevisiae TJ14

BACKGROUND Ethanol production from paper sludge (PS) by simultaneous saccharification and fermentation (SSF) is considered to be the most appropriate way to process PS, as it contains negligible lignin. In this study, SSF was conducted using a cellulase produced from PS by the hypercellulase producer, Acremonium cellulolyticus C-1 for PS saccharification, and a thermotolerant ethanol producer S...
